Comments: | Assay Overview: The purpose of this assay is to determine dose response curves for 28 available out of 28 test compounds that confirmed activity by Relative FRET and Acceptor channel criteria in a Confirmation screen (PubChem AID = 837) which followed a primary screen entitled, "Primary biochemical high-throughput screening assay for inhibitors of the HIV-REV RRE RNA interaction (disruption of protein-RNA interaction)" (PubChem AID = 704). As with the primary and confirmation screens, this assay is based on the ability of the Rev peptide to form a complex with the Rev-Responsive Element (RRE) of double-stranded viral mRNA. Both binding partners have been labeled with appropriate fluorophores in order to monitor their interaction by fluorescence resonance energy transfer (FRET).
